Abstract

Leishmania are digenetic protozoan parasites whose life cycle alternates between two stages: promastigote stage, within the carrier sandfly; and amastigote stage within mammalian host cells. Leishmania’s invasion of the host macrophages causes infections that are either limited to skin or spread to internal organs. In order to cause disease the oarasites must spread within the host. The mechanism through which this takes place is still unknown. In this study, two species causing cutaneous leishmaniasis, GFP-expressing L. mexicana, and L. tropica, were studied for their ability to induce early apoptotic features on infected host cells. L. mexicana caused the highest increase after 24 and 48 hours; 52.7% and 54.56% respectively compared with 28.7 and 34.82% for non-infected THP-1 cells. The effect on apoptosis was found to be infected-cell specific as a higher percentage of apoptotic cells was detected in the sub-population of infected cells. This is the first study were apoptosis and infection are detected at the same time.
