Abstract

Begomoviruses, belonging to the Geminiviridae family, are major plant pathogens that cause severe yield losses in important crops and pose a significant threat to global agriculture. Their ssDNA genome encodes at least three suppressor proteins i.e. AV2/V2 (pre-coat protein), AC2/C2 (transcriptional activator protein) and AC4 protein, which interfere at different steps of host RNA silencing pathway leading to disease development. Like other ]begomoviruses, tomato leaf curl New Delhi virus (ToLCNDV) also contains these three genes. However, suppressor activity of these proteins has not been studied. To evaluate a viral protein's capacity to inhibit RNA silencing, the putative suppressor proteins are co-expressed with a reporter gene, where the suppressor activity is determined by the restoration of reporter gene expression. In the present study, we studied the RNA silencing suppressor activity of AV2/AC2/AC4 proteins of ToLCNDV through GUS reporter assays. Among the three proteins, AV2 showed higher suppressor activity compared to AC2 and AC4 proteins. The suppressor activity of three proteins was then quantified through qRT-PCR. To develop an alternative method of qRT-PCR-based quantification, we describe an image-based quantification assay which is at par with qRT-PCR results.
