Abstract

This study explored the effects of calcium chloride (CaCl2) supplementation during fermentation on the yield, physicochemical properties, and bioactivities of exopolysaccharides (EPS) produced by Lactiplantibacillus plantarum ZGS521. Results showed that CaCl2 significantly upregulated EPS synthesis genes, increasing EPS yield from 467.83 mg/L to 670.82 mg/L. Compared to the conventional fermentation product (LPE), EPS obtained under CaCl2 stress (C-LPE) demonstrated higher carbohydrate and galactose contents, and lower molecular weight. C-LPE enhanced antioxidant capacity of RAW264.7 macrophages by reducing ROS and MDA levels, and increasing SOD and CAT activities, which may be associated with the regulation of Nrf-2/HO-1 signaling pathway. The anti-inflammatory mechanism of C-LPE may involve transcriptional inhibition of TLR4/MyD88 signaling. Additionally, C-LPE showed a protective tendency against intestinal barrier disruption by increasing transepithelial electrical resistance (TEER) values and mRNA expression of tight junction protein in Caco-2 cells. This approach might offer an effective strategy to improve EPS yield and bioactivities in other L. plantarum strains.
