The buccal mucosa has received increased attention in recent years for the delivery of proteins/peptides as an alternative to the currently used parenteral route . Although limited by molecular size, hydrophobicity and low permeability of membranes, thiolated chitosan derivatives have however been identified to deliver proteins/peptides across the buccal mucosa because of their mucoadhesive, penetration enhancing and peptidase inhibition properties . This project aims to develop freeze-dried thiolated chitosan formulations for protein delivery via the buccal mucosa and to determine the effectiveness of cryoprotection and annealing on the stability of the protein drug.
